In 2018, 430 randomly collected feed samples of raw materials and feed mills in farms were analyzed. Samples were tested for aflatoxins (B1, B2, G1, and G2), zearalenone, fumonisins (B1, B2, and B3) and deoxynivalenol by the ELISA Mycotoxin analysis kit (Romer Labs ® ). Results Of 430 feed samples collected, 90.9% were contaminated with deoxynivalenol (table 1). The maximum concentration ...
Depends on the kit assay. Sometimes screening kits may provide false positive reactions. But you can use positive and negative controls along with calibration standards and QC for more accurate results. You can also compare with cross reference method of LC-MS/MS which is more expensive. Hope this will be helpful.
Dear Rena. I have been working with ELISA since the 1980’s, I understand that you may find some false positives, but they are better than false positives and make you recheck the sample with traditional methods. Remember, that in the field, ELISA’S are the most reliable of test kits that we can despond on to either buy grain, feed, other materials. I have worked I Philippines many times and I understand your dilemma, but in reality, you should rely on these tests. There are many ELISA kits in the market and I like to know if you used any of the gold label antibodies rapid tests or you use the conjugate, competition for the antigen bound to the plastic. Best of luck and do rely on these tests.
ELISA is not validated for finished feed, we do not even recommend ELISA for testing of finished feed. Never less, it is the best method for testing raw materials. You can always validate your ELISA equipment and method against reference methods as LC MS/MS or HPLC.
I agree that it needs to be validated with traditional methods as stated in my earlier comments, but there are companies that have validated their tests kits for feeds including raw materials. There are exception that some foreign material may interfere with the test, but again, for field testing is one of the best methods, since there isn’t any traditional method to handle in the field.
