Good review Dr.Khan.It is a commendable job.
The problem with non-conventional vaccines like plasmid DNA vaccines,  synthetic peptide, VLP (virus like particles ) , viral vector vaccines and others of the same genre is that they address only the surface antigens and produce antibodies against a limited antigenic spectrum of the infectious agent.
On the other hand the conventional vaccines, mainly the attenuated live vaccines simulate the infectious process, albeit a low grade non-symptomatic infection, and invoke antibodies to the entire three dimensional conformational protein structure of the surface antigens but also against the various intermediates and precursor proteins for the establishment of IMMUNITY ( not only antibodies ) and prevent the infection from virulent agent.
However continued research on nouveau concepts and adjuvants shall definitely lead to a more robust immunization regime in the future.
