I've being trained as an analytical biochemist and worked on POCT/IVD for years. Currently, we are working on developing p4 panside monitoring kit for dairy farms. We've surveyed quite a few publications for the cutoff value of dairy cows if the cows are not pregnant after AI. So, we develop a protocol to make sure if the cows are not pregnant after AI applying ratio of the progesterone levels. And I like to learn from you experts' comments and suggestion. Our designated plan is the following; 1. get p4 conc on day of AI as D0 2. assay p4 on day 18th or 19th after AI as D18 3. D18/D0>>1 the cows are pregnant 4. D18/D0 approach 1, the cows ain't pregnant, then, the cows submitted for another AI ASAP 5. D18/D0 ratio is obscure, then repeat p4 assay on day 20th as D20 6. D20/D0>>1 the cows are pregnant 7. D20/D0 approach 1, the cows ain't pregnant, then, the cows submitted for another AI ASAP 8. for pregnant cows, one more p4 assay at day 40th~150th to get D40~150 9. D40~150/D0>>1 the cows are pregnant for sure
