I would like to ask about the best way to get the pure nucleotides from yeast? I mean the best manufacture protocol?

Generally speaking, there are three methods: autolysis, enzymatic hydrolysis, acid hydrolysis The autolysis method is used the yeast cell's own enzyme to dissolve the cell wall and release the nutrients in the yeast; while the enzymatic method achieves this effect by adding external enzymes. The advantage of the autolysis method is low cost and can get a good flavor. The advantage of enzymatic hydrolysis is good quality but it's at a high cost. This method is generally used to produce high-end YE products. The advantages of acid hydrolysis are high extract yield and large amounts of free amino acids. However, it has the disadvantage of poor palatability, high salt content, high post-processing cost, and high pollution. So it's rarely used. Nowadays the most often used method is a combination of autolysis-enzymatic hydrolysis process, it's adopted by many manufacturers in China and U.S.A.
