The first multiplexing cytometric bead assay kit for quantitative detection of five mycotoxins
FUNGI-PLEX™ is the first, commercially available multiplexed cytometric bead assay kit for simultaneous detection of mycotoxin contamination in food and feed products.
The first release of the kit allows quantitative detection of five mycotoxins simultaneously:
Ochratoxin A
Aflatoxin B1
Fumonisin B1
T2 Toxin
Zearalenone
Antibodies, specific to the five mycotoxin species are covalently conjugated to six spectrally discrete, *color coded* particles. The coated part ides are used to capture and quantitate the mycotoxin analytes in direct competitive immunoassay Samples are acquired in a flow cytometer and the acquired data are automatically analyzed with the FCAP Array software.
Principles:
- Toxins are extracted from a ground sample with 75 ACN. The extracted samples and a coctail of phycoerythirine (PE-conjugated mycotoxines are mixed and added to the antibody-coated microspheres. Micotoxins from the extracted sample and PE conjugated mycotoxins compete to bind with the antibody coated to the microbeads. The analyte is then measured by detection of the PE fluorescence-emission with flow cytometric analysis.
- The intensity of the PE fluorescence is directly proportional to the amount of bound conjugates and inversely proportional to the concentration of mycotoxins in the sample. Therefore, as the concentration of mycotoxins in the sample increases, the intensity of the PE fluorescence will decrease. The fluorescence intensities of the samples are compared to the corresponding intensities of the kit standards and the sample concentrations are determined.

Versatility. Flexibility
• The matrix effect is low as compared to the ELISA technology, primarily due to the fluorescence based detecting system. Because the matrix effect is low, the FUNGI-PLEX1 technology performs uniformly with a wide range of samples and extraction methods. The kit represents a robust assay relatively tree of interferences for a great variety of agricultural products.

Reproducibility
